Rough PDO prep

DAY 1: Transform & plate out; prep buffers
DAY 2: overnight & autoclave LB for next day
DAY 3: Grow up 8L, pellet (expression gel)
DAY 4: lyse cells, heat purify, histrap, dialyse x3, TEV-digest overnight (purification progression gel)
DAY 5: histrap (w/100mM NaCl), dilute salt out, anion exchange purify, concentrate up (SDS and native gels)
